Cimetidine and renal failure.

چکیده

Introduction Cimetidine is a recently introduced but widely used histamine H2-antagonist. It is known to produce significant increases in plasma creatinine in normal people but the increased values usually remain in the normal range (Burland and Simkins, 1977). In clinical trials a tendency for plasma creatinine to rise, particularly in patients treated with high doses of cimetidine, was found but with continued treatment values generally remained steady or fell (Haggie, Fermont and Wyllie, 1976; Blackwood et al., 1976). McElligott (1978) reported a patient who had a reversible deterioration in renal function during cimetidine therapy and a further case is now reported where a rapid resolution of symptoms and improvement in renal function followed cessation of cimetidine.

A Study on the Effect of Cimetidine on Myoglobinuric Acute Kidney Injury in Male Rats

Introduction: Rhabdomyolysis-induced myoglobinuric acute renal failure accounts for about 10-40% of all cases of acute renal failure (ARF). Iron and Cytochrome P450 are involved in the pathogenesis of myoglobinuric ARF. The Aim of this Study: To explore the effect of cimetidine as a potent cytochrome P450 inhibitor on myoglobinuric ARF in rat. Methods: Twenty four rats were divided into 3 group...

Evaluation the Effect of Cimetidine, Estradiol and Vitamin E on Myoglobinuric Renal Toxicity in Rats

Iron from injured muscles can produce acute renal failure. Cytochrome P 450 is one enzyme that involved in iron-related acute renal failure. Inhibition this enzyme may decrease rhabdomyolysisinduced myoglobinuric nephrotoxicity. The aim of this study was comparison effect of cimetidine, estradiol and vitamin E on myoglobinuric renal toxicity in rats. This study was conducted in 6 groups. Normal...

Down-regulation of rat organic cation transporter rOCT2 by 5/6 nephrectomy.

BACKGROUND In rat kidneys, the organic ion transporters rOCT1, rOCT2, rOAT1 and rOAT3 are considered to mediate the basolateral uptake of various ionic compounds. However, their changes in chronic renal failure (CRF) are poorly understood. The present study examined the renal handling of organic ions and the expression of these transporters under CRF. METHODS 5/6 Nephrectomized rats were used...

Competitive inhibition of the luminal efflux by multidrug and toxin extrusions, but not basolateral uptake by organic cation transporter 2, is the likely mechanism underlying the pharmacokinetic drug-drug interactions caused by cimetidine in the kidney.

Cimetidine, an H₂ receptor antagonist, has been used to investigate the tubular secretion of organic cations in human kidney. We report a systematic comprehensive analysis of the inhibition potency of cimetidine for the influx and efflux transporters of organic cations [human organic cation transporter 1 (hOCT1) and hOCT2 and human multidrug and toxin extrusion 1 (hMATE1) and hMATE2-K, respecti...
